Induction of M2 macrophage Subpopulation by ED-Siglec-9, 2025 - 2029

Summary： This study focuses on the immunomodulatory function of ED-Siglec-9, a soluble ectodomain secreted by dental pulp stem cells (DPSCs), which has been shown to promote M2 macrophage polarization. The research aims to evaluate its role in resolving pulp inflammation and explore its potential as a biologic agent for vital pulp therapy. Both in vitro and in vivo experiments will be conducted. In vitro, THP-1–derived macrophages will be treated with DPSC-conditioned medium (DPSC-CM) and ED-Siglec-9 to assess polarization markers and NF-κB pathway activity. In vivo, a rat model of pulpitis will be used, and ED-Siglec-9 will be delivered via a thermosensitive hydrogel to evaluate inflammatory resolution and macrophage phenotypes. The expected outcome is to identify ED-Siglec-9 as a key immunoregulatory factor that mediates anti-inflammatory effects via NF-κB inhibition, offering a cell-free, clinically translatable approach to regenerative endodontics.
