Establishment of novel oral cancer stem cell model mimicking human oral cancer by reprogramming and insertion of gene mutation, 2019.04 - 2022.03
Summary： To date, no experimental model has been developed that mimics the genetic abnormalities actually found in human oral cancer patients. The gene mutation of TP53, CDKN2A, and FAT1 are the top three gene mutations frequently found in human oral cancer. In this study, we generated TP53 gene-deficient mice and CDHN2A and FAT1 knock-in mice that mimic the gene mutations. We isolated keratinocytes from the tongue of these mice. Currently, we are in the process of establishing an oral cancer stem cell model that mimics human oral cancer by inducing incomplete reprogramming of keratinocytes. We would like to clarify the details of the development and progression mechanism of oral cancer from the analysis using these cells.

Establishment of highly metastatic mice model of oral squamous cell carcinoma, 2017.06 - 2019.03
Summary： Oral squamous cell carcinoma is major type of tumor arising from oral cavity. It is extremely important factor for poor prognosis. However, previous studies are almost using in vitro experiments but not in vivo experiments. In present study, we tried to establish highly metastatic mice model of oral squamous cells carcinoma by using orthotropic transplantation and shRNA library screening. Finally, we could not reach to perform shRNA library screening but established murine oral squamous cell carcinoma cell lines which can apply for in vivo imaging.

Study on the generation of oral cancer stem cell and analysis of its nature, 2017.06 - 2019.03
Summary： Cancer stem cells cause relapse by treatment resistance. Therefore, removal of cancer stem cells is considered to be a useful therapeutic method for preventing metastasis and recurrence. So far, much, the nature and its presence of oral cancer stem cells have not been clarified. In the present study, artificial oral cancer stem cells (iOCS) were generated via constitutively inactivating APC/C by overexpression of Emi1 in oral cancer cells to mimick the cell cycle control of embryonic stem cells. We found that the iOCS cells had stem cell properties such as reduced proliferation, shortened G1 phase and longer S phase in the cell cycle, and increased expression of stem cell markers.

Development of a novel treatment method for xerostomia by dental pulp stem cell-derived humoral factor, 2017.04 - 2020.03
Summary： A model mouse for skin and salivary gland disorder caused by irradiation exhibited significant ulcer formation was observed on the skin. On the other hand, the human dental pulp stem cell culture supernatant-administered group showed a just slight inflammation and skin disorders were significantly reduced at histological level. In the salivary glands irradiation, the acinus contracts and a tissue gap occurs. In the culture supernatant administration group, acinar atrophy was suppressed, and the number of Tunel-positive apoptotic cells was significantly decreased. Furthermore, the cells expressing the cell proliferation marker and the acinar stem cell marker significantly increased in the culture supernatant administration group. From these analysis results, it was suggested that human dental pulp stem cell culture supernatant exerts a regenerating effect on injured salivary glands in addition to a protective effect on salivary gland cells.

Clarification of the mechanism of oral carcinogenesis by the defect of degradation of chromosome passenger complex protein, 2016.04 - 2019.03
Summary： Accurate distribution of replicated chromosomes in cell division is essential for the development of an individual and the inheritance of life to the next generation. The excess or deficiency of the number of chromosomes caused by chromosomal segregation abnormality is called chromosomal instability and is an important step to cause carcinogenesis, but the cause has not been clarified yet. In this study, we focused on Borealin, a component of the chromosome-passenger complex that plays an essential role in controlling chromosome segregation. We clarified its mechanism of degradation and its mechanism of oral carcinogenesis.

Analysis of autoimmunity caused by environmental factors~New pathogenesis of autoimmune disease~, 2016.04 - 2019.03
Summary： Mechanism of the onset of autoimmune disease is so complex that the radical therapy of autoimmune disease has not been established. In this research, the relationship between autoimmunity and environmental factors, such as aging, sex hormone, and metabolism was investigated using animal models for autoimmune disease including Sjogren's syndrome. As molecular pathogenesis associated with aging, abnormal expression of a chemokine receptor on T cells, pathogenic chemokine secretion from resident macrophages in the target organ of autoimmunity, and abnormal expression of a gene in the thymus tissue in the pregnant mice associated with sex hormone were observed using several animal models for autoimmune disease. These results may be useful for development of new clinical application including pathogenesis-based diagnosis and therapy.

Elucidation of novel mechanism to lead DNA over-replication, 2016.04 - 2019.03
Summary： DNA replication is restricted to occur once per cell cycle and its deregulation cause excessive chromosomal numbers called DNA re-replication. The chromosomal abnormality is one of the hallmarks of cancer. So, the understanding of its regulation is important for analyzing cancer biology. In this study, we try to identify novel mechanisms which suppress DNA re-replication by using genome-wide siRNA library. We found that knockdown of each genes did not cause DNA re-replication besides known regulators. Finally, we identified the novel regulators of DNA re-replication by siRNA screening with DNA re-replication inducing reagent.

Analysis of migration factor expressed in dendritic cells during metal allergy development, 2015.04 - 2018.03
Summary： The aim of this study is to clarify the interactions between dendritic cells and keratinocytes during the development of metal allergy. We focused on the interactions of TSLP in keratinocytes and TSLPR in dendritic cells. Expressions of both TSLP and TSLPR in cells stimulated with nickel were increased subsequently. Significant decrease of ear swellings was observed in mice which were injected with TSLPR siRNA in ear skin before the injection of nickel. Therefore, interactions between TSLP in keratinocytes and TSLPR in dendritic cells may play an important role in development of metal allergy and these interactions have a possibility of using for diagnosis or treatment for metal allergy.

Analysis of molecular mechanisms of sensitization phase to develop effective treatments against metal allergy, 2015.04 - 2018.03
Summary： The aim of this study is to clarify the mechanisms of sensitization phase of metal allergy development by analyzing the effect of dental metals upon immune cells. We showed dendritic cells (DC) can uptake nickel directly by using fluorescent particles that recognized nickel in vitro and same phenomenon was observed in skin. Expressions of CD100, MAPkinase, and RhoA in DC stimulated with nickel were increased subsequently. This suggested that these cascades might regulate DC's migration after the uptake of Ni. In addition, expression of plexin B1 and plexin B2 were increased in keratinocytes stimulated with nickel. Therefore, interactions between plexin Bs and CD100 may play an important role in development of sensitization and these interactions have a possibility of using for diagnosis or treatment for metal allergy.

Explore for master regulator of EMT in oral cancer, 2015.04 - 2017.03
Summary： It is known that Epithelial Mesenchymal Transition, EMT is important step in cancer invasion and metastasis. Interestingly, there are spindle cell carcinoma with EMT phenotype known as the subtype of oral squamous cell carcinoma . In this study, we tried to explore the master regulator of EMT by using spindle cell carcinoma cell lines which we previously established. We could not identify the master regulator of EMT but established the spindle cell carcinoma cell line with reporter gene. In future, we try to identify the master regulator of EMT by using this reporter cell line.
